This thesis investigates transcriptional regulation, focusing on DNA–protein interactions as a primary control mechanism of gene expression. Using the bacterial regulators PurR from Escherichia coli and MtrR from Neisseria gonorrhoeae as model systems, a combination of biochemical, biophysical, and structural approaches was employed to examine their DNA binding mechanisms.
