Using neuroanatomical tracing and immunohistochemistry, researchers identified pathways linking the central melanocortin system to brown adipose tissue function. Experiments with MC4R-deficient and pharmacologically treated mice showed that MC4R signaling is necessary for appropriate thermogenic responses to high-fat diets and short-term cold exposure, particularly through regulation of UCP1 expression. These effects appear to occur independently of the hypothalamic-pituitary-thyroid axis. Findings also suggest distinct regulatory mechanisms for diet- versus cold-induced thermogenesis and potential compensatory adaptations over time.
