Although many potent antimalarial therapies exist, the effectiveness of these compounds has been limited by the emergence of drug resistant Plasmodium parasites, the cost of multi-day treatment, and restrictive mechanisms of action, which often fail to protect against the sexual or liver stage parasites that are responsible for disease transmission. A major priority for antimalarial drug development, and this project, is the identification of multi-stage therapies that are broadly effective as treatments, prophylactics, and transmission blocking agents following a single, oral dose. This project focuses specifically on the cytochrome bc1 complex (cyt bc1) as a target for single-dose, multi-stage therapy. Biologically, cyt bc1 is essential for the regeneration of ubiquinone, which plays an essential role in major metabolic processes for P. falciparum, including heme and pyrimidine biosynthesis. 000002781 540__ $$fCC BY 000002781 542__ $$fIn copyright - single owner 000002781 650__ $$aMalaria$$021755 000002781 650__ $$aMitochondria$$022348 000002781 650__ $$aElectron Transport Complex III$$027540 000002781 650__ $$aDrug Therapy$$018086 000002781 691__ $$aSchool of Medicine$$041369 000002781 692__ $$aDepartment of Physiology and Pharmacology$$041442 000002781 7001_ $$aStickles, Allison$$uOregon Health and Science University$$041354 000002781 8564_ $$9119bb5e1-3e8a-46d9-9604-89afc5bd1c28$$s10802813$$uhttps://digitalcollections.ohsu.edu/record/2781/files/3549_etd.pdf$$ePublic$$2c429a1e79333fb31309662caa1e6bdaa$$31 000002781 905__ $$a/rest/prod/w0/89/2b/17/w0892b17j 000002781 909CO $$ooai:digitalcollections.ohsu.edu:2781$$pstudent-work 000002781 956__ $$aGet Accessible Copy$$uhttps://ohsu.libwizard.com/f/requestaccessibledocument 000002781 980__ $$aTheses and Dissertations
