This dissertation investigates structural, regulatory, and physiological roles of CPTI using molecular and mouse models. Mutational analysis of the N‑ and C‑terminal domains of liver CPTI demonstrates compensatory and additive effects on malonyl‑CoA sensitivity, supporting interdomain regulatory interactions. In vivo studies using muscle CPTI knockout and transgenic mouse models reveal that CPTI is essential for development, regulates fatty acid metabolism during fasting, and influences body weight, lipid storage, and susceptibility to diabetes. These findings highlight CPTI as a key regulator of energy metabolism and a potential therapeutic target for metabolic disease. 000000348 540__ $$fCC BY 000000348 542__ $$fIn copyright - single owner 000000348 650__ $$aFasting$$018901 000000348 650__ $$aBody Weight$$015708 000000348 650__ $$aMalonyl Coenzyme A$$021779 000000348 650__ $$aMice$$036842 000000348 650__ $$aMice, Transgenic$$022253 000000348 650__ $$aMetabolic Diseases$$022100 000000348 650__ $$aCarnitine O-Palmitoyltransferase$$016167 000000348 650__ $$aDiabetes Mellitus$$017671 000000348 650__ $$aDiet, High-Fat$$039538 000000348 6531_ $$aglobal gene knockout 000000348 6531_ $$ain vivo overexpression 000000348 6531_ $$amutation activity 000000348 6531_ $$amuscle isoform 000000348 691__ $$aSchool of Medicine$$041369 000000348 692__ $$aDepartment of Science and Engineering$$041447 000000348 7001_ $$aChen, Zhan$$uOregon Health and Science University$$041354 000000348 7201_ $$aWoldegiorgis, Gebretateos$$uOregon Health and Science University$$041354$$7Personal$$eAdvisor 000000348 8564_ $$90a99ae73-d9d4-4b33-8742-a02a79cee2d3$$s984436$$uhttps://digitalcollections.ohsu.edu/record/348/files/349_etd.pdf$$ePublic$$282e3fd1ff0110395d808620f250ee108$$31 000000348 901__ $$a<p>These documents are archival records.
