This work examines the T cell populations and molecular mechanisms that enhance anti‑tumor efficacy in this setting. We demonstrate that durable, potent anti‑tumor responses require the presence of both tumor‑specific CD4⁺ and CD8⁺ T cells in the lymphopenic host. Additionally, we show that T cells deficient in the ubiquitin‑specific protease Ubp43 exhibit superior therapeutic activity compared to wild‑type cells. These findings highlight key cellular components essential for effective adoptive T cell therapy and suggest a strategy to improve clinical outcomes by targeting Ubp43. 000000357 540__ $$fCC BY 000000357 542__ $$fIn copyright - single owner 000000357 650__ $$aImmune System$$020666 000000357 650__ $$aImmunotherapy$$020724 000000357 650__ $$aNeoplasms$$022758 000000357 650__ $$aCD8-Positive T-Lymphocytes$$030782 000000357 650__ $$aAdoptive Transfer$$031483 000000357 650__ $$aMelanoma$$021989 000000357 691__ $$aSchool of Medicine$$041369 000000357 692__ $$aDepartment of Molecular and Medical Genetics$$041428 000000357 7001_ $$aFriedman, Kevin$$uOregon Health and Science University$$041354 000000357 7201_ $$aFox, Bernard$$uOregon Health and Science University$$041354$$7Personal$$eMentor 000000357 8564_ $$9fcc61c66-0cc3-4fff-87e6-656835ba8435$$s1247138$$uhttps://digitalcollections.ohsu.edu/record/357/files/358_etd.pdf$$ePublic$$20f16998530c1ad39ee858b4b0cf002a4$$31 000000357 905__ $$a/rest/prod/5d/86/p0/22/5d86p0224 000000357 909CO $$ooai:digitalcollections.ohsu.edu:357$$pstudent-work 000000357 956__ $$aGet Accessible Copy$$uhttps://ohsu.libwizard.com/f/requestaccessibledocument 000000357 980__ $$aTheses and Dissertations
