We developed and validated 13-color flow cytometry assays on the DxFLEX platform for BCMA and CD19 CAR-T-cells. Methods: Positive samples from MD Anderson Flow Cytometry laboratory (n=10) were analyzed for the comparative study. Both BCMA and CD19 CAR-T assays demonstrated high sensitivity (0.1%), specificity (100%), and excellent accuracy with strong correlation coefficients (R² = 0.99). Compared to normal T-cells, both CAR-T populations expressed significantly higher levels of CD25, CD69, and PD-1. Incorporating immunoregulatory markers could enhance a real-time immune monitoring approach and potentially provide a personalized strategy for managing CAR-T patients.
