Gene expression profiling of OX40‑activated antigen‑specific T cells suggested involvement of the transcription factor c‑Myc and its antagonists Mad4 and Mnt. Using an adoptive transfer model of OVA‑specific DO11.10 T cells in BALB/c mice, we found that OX40 engagement increased T‑cell proliferation and survival. Western blot analyses showed that Mad4, Mnt, and c‑Myc were upregulated early after OX40 stimulation, with OX40 signaling stabilizing Mad4 and Mnt proteins by preventing their degradation. siRNA knockdown of Mad4 and Mnt reduced survival of OX40‑activated T cells, indicating these proteins help counterbalance c‑Myc–driven activation and support memory T‑cell development. 000000536 540__ $$fCC BY 000000536 542__ $$fIn copyright - single owner 000000536 650__ $$aT-Lymphocytes$$026731 000000536 650__ $$aOX40 Ligand$$037329 000000536 650__ $$aRepressor Proteins$$025328 000000536 650__ $$aMemory T Cells$$013784 000000536 650__ $$aGene Expression Profiling$$032769 000000536 650__ $$aGene Expression Profiling$$032769 000000536 650__ $$aProto-Oncogene Proteins c-myc$$029021 000000536 691__ $$aSchool of Medicine$$041369 000000536 692__ $$aDepartment of Cell, Developmental and Cancer Biology$$041399 000000536 7001_ $$aVasilevsky, Nicole$$uOregon Health and Science University$$041354 000000536 7201_ $$aWeinberg, Andy$$uOregon Health and Science University$$041354$$7Personal$$eMentor 000000536 8564_ $$9f701566d-8e0d-42e3-876e-4b5d0b0d093b$$s4767014$$uhttps://digitalcollections.ohsu.edu/record/536/files/537_etd.pdf$$ePublic$$2b071bc0140cb15c8216af241b0f8be42$$31 000000536 905__ $$a/rest/prod/g7/32/d9/03/g732d9039 000000536 909CO $$ooai:digitalcollections.ohsu.edu:536$$pstudent-work 000000536 956__ $$aGet Accessible Copy$$uhttps://ohsu.libwizard.com/f/requestaccessibledocument 000000536 980__ $$aTheses and Dissertations
