This work examines interactions between rhodopsin and its modulators, including arrestins and rhodopsin kinase (RK). We show that both visual and β-arrestin 1 preferentially bind monomeric rhodopsin in nanodiscs, and RK phosphorylates monomeric rhodopsin via a conserved binding region shared with transducin. Fluorescence studies identified two arrestin sites interacting with rhodopsin’s TM6 base. Additionally, initial characterization of CRIP1a, a CB1 receptor modulator, revealed high β-sheet content and stability, with ongoing crystallization efforts. These findings advance understanding of GPCR signal termination and structural regulation. 000000766 540__ $$fCC BY 000000766 542__ $$fIn copyright - single owner 000000766 650__ $$aFluorescence$$019121 000000766 650__ $$aG-Protein-Coupled Receptor Kinase 1$$036928 000000766 650__ $$aRhodopsin$$025463 000000766 650__ $$aCarrier Proteins$$016184 000000766 650__ $$aArrestin$$031593 000000766 650__ $$aReceptors, G-Protein-Coupled$$035314 000000766 691__ $$aSchool of Medicine$$041369 000000766 692__ $$aDepartment of Biochemistry and Molecular Biology$$041396 000000766 7001_ $$aSinha, Abhinav$$uOregon Health and Science University$$041354 000000766 7201_ $$aFarrens, David$$uOregon Health and Science University$$041354$$7Personal$$eAdvisor 000000766 8564_ $$93991d089-f017-4b95-a72b-0d5ac47f1fd9$$s6594130$$uhttps://digitalcollections.ohsu.edu/record/766/files/769_etd.pdf$$ePublic$$2cffb169c22968fbc594087a01a26180e$$31 000000766 905__ $$a/rest/prod/gh/93/gz/55/gh93gz55n 000000766 909CO $$ooai:digitalcollections.ohsu.edu:766$$pstudent-work 000000766 980__ $$aTheses and Dissertations
