Opioid effects are mediated by the μ‑opioid receptor (MOR), whose regulation through desensitization and recovery contributes to tolerance. This study examined agonist‑specific MOR regulation in locus coeruleus neurons using whole‑cell patch‑clamp recordings of GIRK currents. Opioid efficacy correlated with the extent of desensitization and recovery, while distinct agonists produced unique regulatory profiles. Chronic opioid exposure resulted in drug‑specific adaptations in MOR signaling. These findings suggest that opioid tolerance reflects complex, agonist‑dependent cellular mechanisms with important implications for pain management and addiction treatment.
