TY - GEN AB - Acute Myelogenous Leukemia (AML) is an aggressive cancer of the bone marrow that affects both adults and children alike. While many patients achieve remission following induction of chemotherapy, the cancer eventually returns in 40% with increased resistance to cancer drugs. Due to high rates of relapse and drug resistance, the 5-year survival remains less than 25%. Despite mounting evidence implicating EVs in bone marrow dysfunction, more research is needed to fully map the directional transfer of AML-EVs to recipient cells, and determine how their respective cargos enforce pro-leukemic alterations to the bone marrow and extramedullary tissues. In this thesis, I investigate the fate and function of AML-EV in altering the leukemic microenvironment through direct interactions with HSPCs, supportive stroma and cytotoxic T-lymphocytes.
