Novel techniques were developed to purify CB1 and study dynamic structural changes via fluorescence spectroscopy, including the effects of an allosteric ligand. Additional work examined the structure and role of the receptor’s long N-terminus and generated CB1-specific antibodies, one of which is conformationally sensitive and modulates receptor activity. These findings advance understanding of GPCR structure-function relationships and provide tools for probing allosteric modulation and biased signaling in cannabinoid pharmacology. 000000851 540__ $$fCC BY 000000851 542__ $$fIn copyright - single owner 000000851 650__ $$aAllosteric Regulation$$014463 000000851 650__ $$aCannabinoids$$016024 000000851 650__ $$aReceptors, G-Protein-Coupled$$035314 000000851 650__ $$aStructure-Activity Relationship$$026484 000000851 650__ $$aSpectrometry, Fluorescence$$026216 000000851 650__ $$aReceptor, Cannabinoid, CB1$$035348 000000851 691__ $$aSchool of Medicine$$041369 000000851 692__ $$aDepartment of Biochemistry and Molecular Biology$$041396 000000851 7001_ $$aFay, Jonathan$$uOregon Health and Science University$$041354 000000851 7201_ $$aFarrens, David$$uOregon Health and Science University$$041354$$7Personal$$eAdvisor$$eMentor 000000851 8564_ $$9b914a0ab-2a3a-4b63-8d6a-0b10e06dcf03$$s8142163$$uhttps://digitalcollections.ohsu.edu/record/851/files/854_etd.pdf$$ePublic$$2b4441803f695dec02cb909ad9766ea1a$$31 000000851 905__ $$a/rest/prod/zs/25/x8/52/zs25x852h 000000851 909CO $$ooai:digitalcollections.ohsu.edu:851$$pstudent-work 000000851 980__ $$aTheses and Dissertations
