This study examined tumor-specific CD4 T cells in adoptive immunotherapy and their interaction with CD8 T cells in melanoma-bearing lymphopenic mice. Co-transfer of CD4 and CD8 T cells improved therapeutic efficacy, sustained effector CD8 phenotypes, and reduced exhaustion markers compared to CD8-only treatment. Additional experiments in a multi-vaccination model showed that partial CD4 depletion reduced regulatory T cell expansion and enhanced tumor protection. Findings underscore the dual nature of CD4 T cells and highlight strategies to optimize their use in cancer immunotherapy. 000000869 540__ $$fCC BY 000000869 542__ $$fIn copyright - single owner 000000869 650__ $$aMelanoma$$021989 000000869 650__ $$aCancer Vaccines$$031661 000000869 650__ $$aImmunotherapy$$020724 000000869 650__ $$aT-Lymphocytes$$026731 000000869 650__ $$aLeukopenia$$021458 000000869 650__ $$aLymphopenia$$021702 000000869 650__ $$aCD4-Positive T-Lymphocytes$$028453 000000869 650__ $$aT-Lymphocytes, Regulatory$$036362 000000869 650__ $$aImmunotherapy, Adoptive$$028974 000000869 6531_ $$acancer 000000869 691__ $$aSchool of Medicine$$041369 000000869 692__ $$aDepartment of Molecular Microbiology and Immunology$$041429 000000869 7001_ $$aChurch, Sarah$$uOregon Health and Science University$$041354 000000869 7201_ $$aFox, Bernard$$uOregon Health and Science University$$041354$$7Personal$$eAdvisor$$eMentor 000000869 8564_ $$9605a03f3-f926-4f27-ae17-4be41ccc990c$$s2640231$$uhttps://digitalcollections.ohsu.edu/record/869/files/872_etd.pdf$$ePublic$$2ed3e28a1544db1a68839d9c59a5ad2b4$$31 000000869 905__ $$a/rest/prod/90/19/s2/52/9019s252s 000000869 909CO $$ooai:digitalcollections.ohsu.edu:869$$pstudent-work 000000869 956__ $$aGet Accessible Copy$$uhttps://ohsu.libwizard.com/f/requestaccessibledocument 000000869 980__ $$aTheses and Dissertations
