Using optogenetics and whole-cell recordings, we examined opioid sensitivity of GABA inputs to midbrain dopamine neurons from VTA interneurons, nucleus accumbens (NAc), and rostromedial tegmental nucleus (RMTg). RMTg inputs showed strong μ-opioid receptor-mediated inhibition, NAc inputs moderate inhibition, and VTA interneurons minimal inhibition. Striatal inputs exhibited mixed μ- and κ-opioid sensitivity. These findings identify RMTg as a major source of opioid-sensitive inhibition, providing insight into mechanisms underlying opioid-induced dopamine activation. 000000879 540__ $$fCC BY 000000879 542__ $$fIn copyright - single owner 000000879 650__ $$aMethadone$$022132 000000879 650__ $$aMesencephalon$$022077 000000879 650__ $$aVentral Tegmental Area$$030064 000000879 650__ $$aRhodopsin$$025463 000000879 650__ $$agamma-Aminobutyric Acid$$019337 000000879 650__ $$aDopaminergic Neurons$$039537 000000879 650__ $$aReceptors, Opioid, mu$$029979 000000879 6531_ $$amethadone hydrochloride 000000879 691__ $$aSchool of Medicine$$041369 000000879 692__ $$aVollum Institute$$041509 000000879 7001_ $$aMatsui, Aya$$uOregon Health and Science University$$041354 000000879 7201_ $$aWilliams, John$$uOregon Health and Science University$$041354$$7Personal$$eMentor 000000879 8564_ $$9db88d6b4-1ccc-4d6a-8f89-b59075481b78$$s1350189$$uhttps://digitalcollections.ohsu.edu/record/879/files/882_etd.pdf$$ePublic$$2f4a68f89bfa733d30da9c250c0747f53$$31 000000879 905__ $$a/rest/prod/66/82/x3/95/6682x395j 000000879 909CO $$ooai:digitalcollections.ohsu.edu:879$$pstudent-work 000000879 980__ $$aTheses and Dissertations
