This work investigates whether sequential immunization with Env variants derived from HIV-infected individuals who developed bNAbs can guide B cell maturation toward breadth. Using phylogenetic analysis of viral quasispecies and antigenic profiling, we selected Env immunogens and tested DNA-plus-protein co-immunization strategies in rabbits. Results suggest that increasing Env sequence divergence and combining DNA with recombinant gp140 protein enhances neutralization responses compared to protein alone. These findings provide insight into antigenicity-immunogenicity relationships and inform rational design of HIV vaccines aimed at eliciting bNAbs. 000000884 540__ $$fCC BY 000000884 542__ $$fIn copyright - single owner 000000884 650__ $$aAIDS Vaccines$$029526 000000884 650__ $$aImmunoglobulins$$020695 000000884 650__ $$aRabbits$$025058 000000884 650__ $$aAntibodies$$014849 000000884 650__ $$aHIV Infections$$028555 000000884 650__ $$aVaccine Development$$013568 000000884 650__ $$aQuasispecies$$012396 000000884 650__ $$aBroadly Neutralizing Antibodies$$013185 000000884 650__ $$aHIV$$020258 000000884 650__ $$aenv Gene Products, Human Immunodeficiency Virus$$037739 000000884 650__ $$aAntibodies, Neutralizing$$038855 000000884 691__ $$aSchool of Medicine$$041369 000000884 692__ $$aDepartment of Molecular Microbiology and Immunology$$041429 000000884 7001_ $$aPissani, Franco$$uOregon Health and Science University$$041354 000000884 7201_ $$aHaigwood, Nancy$$uOregon Health and Science University$$041354$$7Personal$$eAdvisor 000000884 8564_ $$91382b364-4672-43da-9130-d1e2ed012423$$s3035729$$uhttps://digitalcollections.ohsu.edu/record/884/files/887_etd.pdf$$ePublic$$23dfc1ee1ba2c499d9c358608c8878aa3$$31 000000884 905__ $$a/rest/prod/p8/41/8n/25/p8418n25z 000000884 909CO $$ooai:digitalcollections.ohsu.edu:884$$pstudent-work 000000884 956__ $$aGet Accessible Copy$$uhttps://ohsu.libwizard.com/f/requestaccessibledocument 000000884 980__ $$aTheses and Dissertations
