This thesis leverages novel single-cell epigenomic and proteomic approaches to aid in the identification of precise subtypes of prostate tumors that are organ-confined at the time of diagnosis. We first utilize cyclic immunofluorescent imaging to profile the spatial heterogeneity of prostate tumors and uncover risk-level and Gleason Pattern specific cell types and cellular interactions. Next, we perform single-cell ATAC sequencing of the same tumors and identify unique genomic regions associated with high-risk tumors and immune response signatures associated with low-risk tumors. Finally, we develop assays that enable multiplex imaging of biomarkers isolated from low-volume liquid biopsy samples. Our work uncovers unique cellular and molecular markers, which hold the potential to become novel prognostic factors, and presents a multiplex liquid biopsy approach for the detection of prostate cancer for precise treatment strategies. 000009380 542__ $$fIn copyright - single owner 000009380 650__ $$aProstatic Neoplasms$$024731 000009380 650__ $$aProstate-Specific Antigen$$029960 000009380 650__ $$aLiquid Biopsy$$012229 000009380 650__ $$aImmunity$$020668 000009380 650__ $$aGenomics$$033016 000009380 691__ $$aSchool of Medicine$$041369 000009380 692__ $$aDepartment of Biomedical Engineering$$041397 000009380 7001_ $$aSayar, Zeynep 000009380 8564_ $$9fbe526d5-fbad-4c49-b9ee-cbdfe0ec43df$$s10809815$$uhttps://digitalcollections.ohsu.edu/record/9380/files/Sayar.Zeynep.2021.pdf 000009380 905__ $$a/rest/prod/n5/83/xv/75/n583xv754 000009380 909CO $$ooai:digitalcollections.ohsu.edu:9380$$pstudent-work 000009380 980__ $$aTheses and Dissertations
