This work investigates the molecular interactions critical to the assembly and maturation of human immunodeficiency virus type 1 (HIV‑1). The production of infectious virions requires precisely coordinated protein–protein, protein–membrane, and protein–nucleic acid interactions. In particular, the PrGag and PrGag‑Pol polyproteins must be correctly targeted to cellular assembly sites, interact in a regulated manner, and bud from the host cell. Subsequent proteolytic processing of these polyproteins is essential for virion maturation and infectivity.
